The NRTN-related gene GFRAL is a novel tumor suppressor gene in human oral squamous cell carcinoma.
Glial cell line-derived neurotrophic factor (GDNF) family receptor alpha-like (GFRAL) belongs to the GDNF family receptor (GFR)-alpha family and is a receptor for Neurturin (NRTN), a member of the GDNF family. It has been reported that NRTN/GFRAL signaling inhibits cell migration and invasion in normal epithelial cells. However, the significance of GFRAL in human oral squamous cell carcinoma (OSCC) remains unclear. In this study, we found that the mRNA and protein levels of GFRAL were decreased in human OSCC samples compared with paired non-tumor tissues. Then, we established cell lines from OSCC (HSC-2, HSG, Ca9-22 and HSC-3) by transfecting GFRAL and found that GFRAL inhibited cell proliferation and migration in vitro and in vivo. Moreover, GFRAL could induce cell-cycle arrest in the S-phase and apoptosis of OSCC cells. More importantly, HSC-2 cells in which GFRAL was stably over-expressed showed decreased tumorigenicity in mice.
