The ability of the SCP2 isoforms to transport cholesterol has been studied using indirect techniques, such as measuring steroidogenesis, to examine possible protein-steroid interactions. We have used an antibody raised against a peptide from the extracellular domain of the 180 kDa SCP2 (SCP2-ab) to test for possible sterol binding, and to examine for sterol binding activity in the PM vesicles. SCP2-ab binding was found to be sigmoidal in concentration and was saturable at a molar ratio of 2:1. Molar ratios of 10:1, 20:1 and 60:1 of sterol to SCP2-ab caused significant and dose-dependent inhibition of SCP2-ab binding, suggesting that the sterol binding site in SCP2-ab is hydrophobic in nature and the binding site is greater than 25 A from the protein surface. The 120-kDa SCP2 isoform (SCP2-b) contains a Ser/Thr cluster (SST) located in the N-terminus, which is phosphorylated in response to P13K activation. It has been suggested that the SST cluster modifies SCP2-b function. However, SCP2-b, but not SCP2-ab, was immunoprecipitated by a phospho-SST antibody, suggesting that SCP2-b may be a substrate for P13K-mediated phosphorylation.
