The PPi:colominic acid monomer association constant at a given pH is a function of the PPi concentration. We show that the apparent binding constant increases with pH. For example, at pH 7.0 the apparent association constant of a 30 mM solution of PPi is 2.00 x 10(3) M-1 at 15 degrees C while that of a 60 mM solution of PPi is 2.43 x 10(4) M-1 at 25 degrees C. The relative increase in the apparent association constant with increasing pH can be described by a simple two-state model in which the free colominic acid monomer adopts the low-energy conformation at high pH and the high-energy conformation at low pH.
